Background: Metastatic disease caused by prostate cancer (PCa) is the leading cause of PCa-related death. RNA long non-protein-coding may have a significant cellular functions. Plasmasitoma variant translocation 1 (PVT1), long non-coding RNA genes encoded by human PVT1, is an oncogene, which can set up multiple tumor-associated genes. In PCa, function and PVT1 mechanism is not clear. NOP2 being pursued as a prognostic marker for cancer aggressiveness, which promotes the growth of mouse fibroblast and tumor formation. Basically, nothing is known about the specific interaction between PVT1 and NOP2.
Methods: 190 pairs of PCa tissue and adjacent normal tissue were collected and RNA sequencing was used to identify differential lncRNAs. Real-time quantitative real-time PCR (RT-qPCR) confirmed these results and the relationship of gene regulation. PVT1 lentiviral vectors are used to alter and genes to analyze their effects on the development of PCa. Transwell migration and invasion assays were performed to test the ability of metastasis. Biofunction of PVT1 and NOP2 confirmed in vitro and in vivo.
Results: In this study, we report that long noncoding RNA-PVT1 is upregulated in PCa tissue promoted metastasis and PCa cell migration in vitro and their metastasis in vivo. High levels were significantly downregulated PVT1 tumor suppressor microRNAs (miRNAs), such as miR-15b-5p, miR-27a-3p, miR-143-3p, and miR-627-5p, levels in metastatic tissues are low compared with those in non-metastatic tissues. In vitro and in vivo, PVT1 promote PCa metastasis by targeting miRNAs. In addition, expression levels associated with the expression of positive PVT1 NOP2, cancer metastasis-associated protein. We show that promoted the invasion and migration NOP2 PCa. For certain mechanisms, correlation analysis showed that PVT1 promoted metastasis to up-regulate NOP2.
Conclusion: Overall, our results indicate that PVT1 acts as an inducer of PCa metastasis by targeting miRNAs, thus increasing NOP2. This axis may have diagnostic and therapeutic potential for advanced PCa.

Pancreatic cancer is a malignant tumor of the digestive tract, rendering it difficult to make an accurate diagnosis. 5-year survival rate for pancreatic cancer is <1%, and surgical resection is rarely proven to be effective. Therefore, the identification of more effective methods for early detection of pancreatic cancer is an urgent need. This study aims to explore key genes and microRNAs (miRNAs) associated with the pathogenesis of pancreatic cancer. public databases were searched, and the integrated data from The Cancer Genome Atlas and Gene Expression Omnibus database, which leads to the identification of 23 miRNAs that are expressed differently (DE-miRNA).
Four of DE-regulated miRNAs (miR-892b-HSA, HSA-miR-194-2, miR-200a-HSA and HSA-miR-194-1), while the DE-19 downregulated miRNA (hsa- Mirza 424, HSA-mir-191, mir-484-HSA, HSA-miR-142, miR-15b-HSA, HSA-miR-450A-1, HSA-mir-423, mir-126-HSA, hsa- Mirza 505, HSA-miR-16-1, HSA-miR-342, miR-130A-HSA, HSA-miR-3613, HSA-miR-450A-2, HSA-miR-26b, miR-451-HSA, HSA- miR-19b-2, HSA-miR-106A and HSA-miR-503) were identified using a cut-off criteria of P <0.05 and | log 2FC |> 1.0.
