A Biomarker Panel of Radiation-Upregulated miRNA as Signature for Ionizing Radiation Exposure
Teer radiation causes serious injury to the human body and has an old impact on health. It is important to find optimal biomarkers for the initial fast playback of individuals exposed. A series of Mirnas signatures have been developed as new biomarkers for diagnosis, survival, and prognostic predictions of cancer. Here, we have identified the MIRNA profile that can induce ionization radiation through microarray analysis. The biological function is predicted to the top six mirnas regulated by 4 GY γ-Rays: MIR-1246, MIR-1307-3P, MIR-3197, MIR-4267, MIR-5096 and MIR-7641. Mirna-gen network and network analysis of the target genes revealed that DNAH3 was a target gene associated with all six MIRNAS. Golgb1 is related to 4 MIRNAS and 26 other genes targeted by 3 MIRNAS.
Fifteen Mirnas upregulation is verified further at 4 hours and 24 hours after 0 to 10 gy irradiation in human AHH-1 Lymphoblastoid cells, and some show increased dose increases. Six Mirnas, including MIR-145, MIR-663, MIR-1273G-3P, MIR-6090, MIR-6727-5P and MIR-7641, validated to be a dose-dependently regulated at 4 hours or 24 hours post-irradiation in good AHH -1 and human peripheral blood lymphocytes are irradiated ex vivo. This Six-Mirna signature displays the advantage as radiation biomarkers for screening translation applications and exposed individual radiation assessments.
Melatonin and MIRNA regulation: novel target therapy for cancer and not warning
MIRNAS, non-non-job RNA with important diagnostic and prognostic capabilities, playing the role of essential therapy in various human diseases. Biomarkers are involved in several biological mechanisms and are responsible for regulations of various expression genes in cells. Mirna-based therapy has shown a very bright future in the case of clinical intervention. Melatonin, the main product of the pineal gland, is a multifunctional neurohormone with many therapeutic potential in human diseases.
Melatonin is able to manage MIRNAS in various pathologies such as malignant and nonmalignant, which can be considered as a targeted type of therapy. Here, this review discusses the possibility of melatonin therapeutic utilities for MIRNA regulations in various pathological conditions.
Hepatoprotective effect of ginkgo biloba extract against hepatotoxicity induced by metotrexation through targeting stat3 / mirna-21 axis
The use of the Methotrexate (MTX) chemotherapy agent is associated with hepatotoxicity which minimizes its clinical use. Ginkgo Biloba (GBE) extract was used previously to relieve mtx-induced liver injury through its antioxidant activity. This work was conducted to explain other molecular hepatoprotective mechanisms from GBE checking the IL-6 / Stat3 path next to the MIRNA-21 expression in the liver tissue. Sprague Dawley Rats allocated into four groups: normal control (NC); Control ginkgo biloba extract (GBEC); methotrexate (MTX); and ginkgo biloba extract and metotrexate group (GBE + MTX). GBE is managed orally 60 mg / kg / day for 10 days while the MTX is injected intraperitone with 20 mg / kg the following day 5.
After the experiment, serum is separated for determining the enzyme liver while the liver tissue is collected for biochemical exams and histopathology. MTX induces the altitude marked in liver enzymes, hepatic IL-6, and HGF MRNA expression, phospho-stat3 / stat3 ratio, and mirna-21 liver expression when compared to the NC group. Heart injury is observed histopathologically after MTX. The GBE government reverses these biochemical changes and increases liver histopathology. The GBE hepatoprotective mechanism for MTX induced hepatotoxicity through the modulation of the IL-6 / Stat3 signal path and downregulation of the MIRNA-21 liver expression was reported for the first time.
Mirna integrated analysis and gene expression changed in response to obesogenic diets to explore the impact of transgeneration supplementation with Omega 3 fatty acids 3
Insulin resistance reduces insulin ability to inhibit hepatic gluconeogenesis, key steps in the development of metabolic syndrome. Metabolic changes, fat accumulation, and fibrosis in the liver are closely related and contribute to the development of comorbidity, such as hypertension, type 2 diabetes, or cancer. Omega 3 (N-3) polyunsaturated fatty acids, such as eicosapentaenoic acid (EPA), are identified as a positive regulator share of insulin sensitivity in vitro and on animal models. In the current study, we explore the effects of transgeneration supplementation with EPA in mice exposed to obesogenic diets in Microrna (MIRNAS) regulations and gene expression at the liver using throughput techniques.
We apply the biology approach to a comprehensive molecular system, combining statistical tools, such as pipe analysis of microrna regulators and boolean modeling to integrate this biochemical process. We show that the molecular adaptation is mediated by EPA, which leads to the inhibition of MIR-34A-5P, the negative regulator of the IRS2 as the main rules that lead to inhibition of gluconeogenesis by insulin during fasting transition. Data integration Omics provides a greater biological insight and a better understanding of the relationship between biological variables. Such an approach might be useful to get hypotheses driven by innovative data and for the discovery of mechanistic mechanistic links-biochemicals.
Biomarkers for suicidal behavior: MIRNAS and their potential for diagnostics through liquid biopsy – systematic reviews
Given that around 70% of MIRNAs in the body are neurons, we assess critically the current study of MIRNAS and suicidal behavior. Material & Method: To determine the role of MIRNNA in suicide, we are looking for studies of extracellular vesicles (exosom) because MIRNAS is specifically enriched in the exosomes. MIRNAS also has an important physiological role, and they can pass the blood-brain barrier and participate in cell-cell communication with nearby and far cells.
